Biological effects of contaminants: Assessing DNA damage in marine species through single-cell alkaline gel electrophoresis (comet) assay. ArchiMer
Bean, Tp; Akcha, Farida.
Single cell gel electrophoresis (SCGE or comet) assay allows quantification of DNA damage in individual cells and is an ideal tool for use within biological monitoring programmes. Comet assay can be used on a range of cell types including somatic, reproductive (gametes) or circulatory cells in many different species including both marine bivalves and flatfish. The assay can be employed with simple equipment available in most laboratories, is sensitive to environmentally relevant levels of DNA damage (Frenzilli and Lyons, 2009), accurately demonstrates a linear dose response to exposure (Collins et al., 1996), and can be adapted for use on most nucleated cell types. This document concentrates on the simplest and most repeatable method of comet assay in...

Biological Effects Produced by Intense Pulsed Light (Xe-Cl) on the Cartilage of the Tongue Chick Embryo Using Various Filters International Journal of Morphology
Avila,R. E; Samar,M. E; Juri,H; Femopase,G. A; Hidalgo,M; Ferrero,J. C; Rinaldi,C; Fonseca,I; Juri,G.
The Laser used correctly in the medical practice offers clear advantages compared with traditional therapies. The improvement and even the elimination of many significant skin lesions can be achieved with reduced risks to patients. However, it is important to keep security measures and understand the possible effects on an experimental model. The chick embryo is a good model to evaluate the direct effects of non-ionizing radiation for its easy handling and availability. The purpose of this communication is to show our histological findings in organs of the chick embryo with and without protective barrier to be subjected to radiation excimer. We used the following issuers: intense pulsed light (excimer Xe-Cl laser of 308 nm wavelength). It was irradiated...

How can we quantify impacts of contaminants in marine ecosystems? The ICON project ArchiMer
Hylland, Ketil; Burgeot, Thierry; Martinez-gomez, Concepcion; Lang, Thomas; Robinson, Craig D.; Svavarsson, Jorundur; Thain, John E.; Vethaak, A. Dick; Gubbins, Matthew J..
An international workshop on marine integrated contaminant monitoring (ICON) was organised to test a framework on integrated environmental assessment and simultaneously assess the status of selected European marine areas. Biota and sediment were sampled in selected estuarine, inshore and offshore locations encompassing marine habitats from Iceland to the Spanish Mediterranean. The outcome of the ICON project is reported in this special issue as method-oriented papers addressing chemical analyses, PAH metabolites, oxidative stress, biotransformation, lysosomal membrane stability, genotoxicity, disease in fish, and sediment assessment, as well as papers assessing specific areas. This paper provides a background and introduction to the ICON project, by...

Integrated chemical and biological assessment of contaminant impacts in selected european coastal and offshore marine areas ArchiMer
Hylland, Ketil; Robinson, Craig D.; Burgeot, Thierry; Martinez-gomez, Concepcion; Lang, Thomas; Svavarsson, Jorundur; Thain, John E.; Vethaak, A. Dick; Gubbins, Mattew J..
This paper reports a full assessment of results from ICON, an international workshop on marine integrated contaminant monitoring, encompassing different matrices (sediment, fish, mussels, gastropods), areas (Iceland, North Sea, Baltic, Wadden Sea, Seine estuary and the western Mediterranean) and endpoints (chemical analyses, biological effects). ICON has demonstrated the use of a framework for integrated contaminant assessment on European coastal and offshore areas. The assessment showed that chemical contamination did not always correspond with biological effects, indicating that both are required. The framework can be used to develop assessments for EU directives. If a 95% target were to be used as a regional indicator of MSFD GES, Iceland and offshore...

Is nuclear medicine really safe? BABT
In nuclear medicine, radiation absorbed dose estimates calculated by standard models at the whole body or organ are very low. At cellular level, however, the heterogeneity of radionuclide distributions of radiation dose patterns may be significant. We present here absorbed doses at cellular level and evaluate their possible impact on the usually assumed risk/benefit relationships in nuclear medicine studies. The absorbed dose values calculated are surprisingly high, and are difficult to interpret. In the present study, we show calculated doses at the cellular level and discuss possible biological consequences, for two radiopharmaceuticals labelled with technetium-99m: human serum albumin microspheres used for pulmonary scintigrapies and HMPAO used to...

Maurice, Manuelle.
In this report we present the main scientific orientations taken until now in France and under the world conceming marine ecotoxicology. We discuss successively the state of the environment in France, the toxicity of pollutants on marine organisms, the monitoring of pollutants and the technics used to measure the biological effects on marine ecosystems.

Micropropagation, antinociceptive and antioxidant activities of extracts of Verbena litoralis Kunth (Verbenaceae) Anais da ABC (AABC)
Braga,Virgínia F.; Mendes,Giselle C.; Oliveira,Raphael T.R.; Soares,Carla Q.G.; Resende,Cristiano F.; Pinto,Leandro C.; Santana,Reinaldo de; Viccini,Lyderson F.; Raposo,Nádia R.B.; Peixoto,Paulo H.P..
This work describes an efficient micropropagation protocol for Verbena litoralis and the study of the antinociceptive and antioxidant activities in extracts of this species. For the establishment in vitro, surface-sterilization procedures and PVPP showed high efficiency in fungal-bacterial contamination and phenol oxidation controls. Nodal segments cultivation in MS medium supplemented with 6-benzyladenine (7.5 µM)/α-naphthaleneacetic acid (NAA; 0.005 µM) induced multiple shoots. Elongated shoots were rooted with IAA (0.2 µM). Acclimatization rates were elevated and the plants showed the typical features of this species. The hexanic fraction (HF) of powdered leaves presented a radical scavenging activity with IC50 = 169.3 µg mL-1. HF showed a non-dose...
